[spa] La diabesidad es un estado patológico que se caracteriza por la interacción metabólica entre la
obesidad y la diabetes mellitus tipo 2, el cual puede alterar la función endocrina del tejido adiposo
blanco. Dados los recientes estudios en los que se demuestra la existencia de un dimorfismo sexual
relacionado con cambios en el secretoma del tejido adiposo gonadal de ratas hembra ZDF debido a la
acción de los estrógenos, fue realizado un análisis de la expresión de determinadas adipoquinas para
ese mismo tejido en un modelo animal de ratas macho ZDF. Dicho modelo de diabesidad se
caracteriza por un incremento del peso y la glucemia de los animales como resultado de una
mutación en el receptor de la leptina.
Los resultados obtenidos en este estudio describen que se produce una mayor expresión de las
adipoquinas PAI-1, IL-6, CD68, MIP-1α, TNF-α y leptina en el tejido adiposo gonadal de ratas macho
diabésicas. Este aumento en la expresión se corresponde a un perfil secretor alterado debido a la
expansión del tejido adiposo, el cual está relacionado con los procesos patológicos que están
asociados a la diabesidad.; [eng] Diabesity is a pathological state characterized by the metabolic interaction between obesity and type
2 diabetes mellitus, which can alter the endocrine function of white adipose tissue. Given the recent
studies that demonstrate the existence of a sexual dimorphism related to changes in the secretome
of the gonadal adipose tissue of female ZDF rats due to the action of estrogens, an analysis of the
expression of certain adipokines for the same was performed. Said model of diabesity is
characterized by an increase in the weight and blood glucose of the animals as a result of a mutation
in the leptin receptor.
The results obtained in this study describe a greater expression of the adipokines PAI-1, IL-6, CD68,
MIP-1α, TNF-α and leptin in the gonadal adipose tissue of diabetic male rats. This increase in
expression corresponds to an altered secretory profile due to the expansion of adipose tissue, which
is related to the pathological processes that are associated with diabesity.

[eng] During last decade cancer therapies have undergone constant progress, as reflected in
the development of novel histological methods for diagnosis, personalized chemotherapy and
targeted therapy. Despite progress made in colorectal cancer (CRC) field, little can be done to
improve overall survival, even on early CRC stages (II-III), besides surgical resection in
combination with chemotherapy. Therefore, new biomarkers for early detection of the disease,
more innovative methods for patient stratification and the development of new
immunotherapeutic treatments become essential. Importantly, several studies have shown that
the tumor microenvironment plays a crucial role in CRC progression. In this context, this study
aimed to lay the groundwork for the first characterization of the membrane lipid fingerprint of
sorted immune cells, which eventually may infiltrate the tumor. For that end, our purpose was
to establish the lipidome of each cell type using MALDI-IMS technology, which exhibits a much
higher sensitivity compared to other MS technologies. Unexpectedly, this approach could not
be directly applied in sorted cells, as the spectra obtained were lacking the most relevant peaks.
Consequently, the study aims was reevaluated and it was focused on investigating the reason of
such failure and on setting up a successful procedure. The latter is essential for the optimal
development of one of the current research lines of the group. Thus, the procedure described
here should allow us characterizing the lipidome of any type of immune cells, even for rare
populations, of which there is no information available yet. The thorough description of immune
cells lipidome will also help for a better comprehension of the complex network of interactions
prevailing within the tumor microenvironment.

[spa] Los gliomas son tumores que se originan a partir de las células madre de la neuroglia o de las
células progenitoras, siendo el glioblastoma (GBM) el tumor cerebral primario más común y
agresivo. La terapia convencional para el GBM consiste en una resección tumoral seguida de
radioterapia y quimioterapia con temozolomida (TMZ), no obstante, los pacientes acaban siendo
resistentes debido principalmente a una subpoblación de células denominadas Glioma Stem
Cells (GSC). Actualmente, se apuesta por la reutilización de drogas (drug repositioning), una
estrategia terapéutica más segura y económica. En esta línea, el objetivo principal del presente
estudio es evaluar el potencial coadyuvante de las fenotiazinas antipsicóticas (trifluoperazina
(TFP) y tioridazina (TRD)), fármacos utilizados en el tratamiento de la esquizofrenia, en líneas
celulares de GBM (U87MG, LN229 y U251). Asimismo, estudiar el papel del supresor tumoral
FOXO3a en respuesta a las fenotiazinas, ya que resultados previos del grupo de investigación
demuestran que inhiben su exportación nuclear. El análisis isobolográfico determinó que de las
9 combinaciones probadas (TMZ:TFP y TMZ:TRD), la 1:1 y la 3:1 eran siempre sinérgicas en
U87MG y en U251. Por otro lado, el estudio de clonogenicidad reveló que, el efecto a largo plazo
de TRD no era permanente ya que las células no perdían la capacidad de formar colonias.
Finalmente, se demostró claramente que las fenotiazinas antipsicóticas favorecían la
importación nuclear de FOXO3a y, por consiguiente, su activación. Todos estos resultados son
muy prometedores y se podrían llegar a sentar las bases de un ensayo clínico.; [eng] Gliomas are tumors that derive from neuroglia stem or progenitor cells, among these
glioblastoma (GBM) is the most common and aggressive primary brain tumor. Conventional
therapy for GBM consists of tumor resection followed by radiotherapy and chemotherapy with
temozolomide (TMZ), however, patients end up being resistant, mainly, due to a subpopulation
of cells called Glioma Stem Cells (GSC). The need for more effective anti-cancer drugs has
sparked a growing interest for drug repositioning, a safer and cheaper therapeutic strategy. In
this line, the main objective of the present study is to evaluate the adjuvant potential of
antipsychotic phenothiazines (trifluoperazine (TFP) and thioridazine (TRD)), drugs used in the
treatment of schizophrenia, in GBM cell lines (U87MG, LN229 and U251). In addition, we aimed
to study the role of the tumor suppressor FOXO3a in response to phenothiazines, as previous
results of the research group demonstrate that these drugs inhibit FOXO3a nuclear export. First,
the isobiolographic analysis determined that two of the nine ratios tested (TMZ: TFP and TMZ:
TRD) were always synergistic in U87MG and in U251 cells. Secondly, the clonogenic assay
revealed that, the long term effect of phenothiazines was not permanent because the cancer
cells did not lose the capacity to form colonies. Finally, it was clearly demonstrated that
phenothiazines induced the nuclear import of FOXO3a and, consequently, its activation. These
results are very promising and the basis of a clinical trial could be established.

[eng] Brain metastasis is a clinically devastating process that occurs in approximately 10-20% of cancer
patients. To properly colonize the brain, cancer cells need to penetrate the blood-brain barrier (BBB),
a highly specialized vasculature with strong intercellular junctions that isolates and protects the brain.
A better understanding of the molecular mechanisms of the extravassation through the BBB is crucial
to be able to find new clinical targets. In this master’s thesis we wanted to characterize brain-tropic
breast metastatic cells migration through the blood-brain barrier using an in vitro model that mimics
some of the BBB properties. We demonstrated that the endothelial cells of the in vitro BBB model
express VE-cadherin and form adherens junctions, and also that the BBB model produces a decrease
in permeability through the transwell. Then, we wanted to characterize brain tropic metastatic cancer
cells (MDA-MB-231 BrM) by studying its adhesion on an endothelial monolayer and transmigration
capacity. We concluded that these brain tropic cancer cells have a higher capacity of adhesion and
transmigration than the parental ones. The validation of this BBB model will allow us to use it to
identify key regulators of cancer cells extravasation to the brain during the metastatic cascade.; [spa] La metástasis cerebral es un proceso devastador que se desarrolla, aproximadamente, en el 10-20%
de los pacientes con cáncer. Para poder llegar al cerebro, las células metastásicas deben penetrar la
barrera hematoencefálica, una vasculatura altamente especializada con fuertes uniones intercelulares
que aísla y protege el cerebro. Un mayor conocimiento de los mecanismos moleculares que subyacen
al proceso de la extravasación a través de la barrera hematoencefálica es crucial para poder encontrar
nuevas dianas terapéuticas. En este trabajo de fin de máster se ha caracterizado la transmigración de
células de cáncer de mama metastásicas con tropismo hacia el cerebro mediante el establecimiento
de un modelo de barrera hematoencefálica. Pudimos comprobar que las células endoteliales que
constituyen el modelo de barrera hematoencefálica expresan VE-cadherina y, por tanto, presentan
uniones adherentes entre sí, además de demostrar una importante bajada de permeabilidad con este
modelo. Después, se caracterizó una línea celular de cáncer de mama con tropismo hacia el cerebro
(MDA-MB-231 BrM) mediante el estudio de su capacida de adhesión sobre una monocapa de céluals
endoteliales, así como de la transmigración a través del modelo de barrera hematoencefálica,
pudiendo concluir que estas células presentan una mayor capacidad de adhesión y transmigración que
las células parentales. La validación de este modelo de estudio permitirá su uso para identificar a
reguladores clave implicados en la extravasación de las células tumorales a través de la barrera
hematoencefálica durante la cascada metastásica.
