[spa] El fenotipo MONW (metabolically obese but normal weight) hace referencia a aquellos
individuos que presentan un peso normal pero que están alterados a nivel metabólico. A día
de hoy existen pocos estudios focalizados en las alteraciones en el control de la ingesta de
estos individuos.
En el presente trabajo quisimos analizar las posibles alteraciones relacionadas con la
homeostasis energética y el control de la ingesta asociadas al fenotipo MONW. Para ello
analizamos la expresión de genes clave mediante RT-PCR a tiempo real en el hipotálamo de
ratas Wistar macho alimentadas con diferentes dietas ricas en grasa: 45% de grasa (HF45) o
60% de grasa (HF60%), ambas en cantidades isocalóricas respecto al grupo control.
Los resultados mostraron una disminución significativa de la expresión de Npy, acompañada
de una menor expresión de Socs3 en el grupo HF45, lo que sugiere que estos animales
intentan contrarrestar el gran aporte de energía en forma de grasa inhibiendo la sensación de
hambre y aumentado la sensación de saciedad. Sin embargo, el grupo HF60 presentó una
tendencia a disminuir la expresión del gen que codifica para el péptido anorexigénico CART,
lo que se tradujo en un aumento del ratio Agrp/Cart, sugiriendo un aumento de la sensación
de hambre en estos animales.
En conclusión, el grupo HF45 estaría activando mecanismos compensatorios de inhibición de
la ingesta frente al elevado aporte de grasa, mientras que las alteraciones observadas en el
grupo HF60 sugieren una disminución de la sensación de saciedad. |

[eng] The MONW (metabolically obese but normal weight) phenotype refers to those individuals
that have a normal weight but are altered metabolically. As far as we know, there are a low
number of studies specially focused on the food intake alterations of MONW.
In this study we wanted to analyse the possible changes related to energy homeostasis and
control of intake associated with the phenotype MONW. In this sense, we analysed the
expression of key genes by RT-PCR in real time in the hypothalamus of male Wistar rats fed
with different high fat diets: with 45% of fat (HF45) or 60% of fat (HF60), both isocaloric
amounts compared to the control group.
The results showed a clear decrease on Npy and Socs3 gene expression on HF45 animals,
suggesting that the animals try to counteract the high amount of ingested fat, reducing hunger
sensation and increasing satiety sensation. However, HF60 group revealed a tendency to
decrease the expression of the gene which codes for the CART anorexigenic peptide, which
was related with an increase of Agrp/Cart ratio, suggesting an increase on hunger sensation of
these animals.
In conclusion, HF45 group would activate compensatory mechanisms of inhibition of intake
to counteract the high fat intake, while the HF60 group loss of satiety is suggested. |
