[spa] En las últimas décadas se ha producido un incremento en la esperanza de vida, lo que ha supuesto un aumento de la incidencia de las enfermedades asociadas al envejecimiento. La teoría del estrés oxidativo es una de las teorías más aceptadas para explicar las causas del envejecimiento, así como un incremento de la respuesta inflamatoria en la que el estrés oxidativo podría estar involucrado. El fenotipo inflamatorio crónico característico del envejecimiento contribuye al declive de las funciones fisiológicas, siendo este uno de los principales factores de riesgo para diferentes patologías como son las enfermedades neurodegenerativas. Actualmente el envejecimiento es objeto de investigación para determinar las bases moleculares que lo producen y modulan, con el fin de poder prevenir o retrasar sus consecuencias. Entre las terapias existentes cabe destacar la realización de ejercicio físico y administración de antioxidantes por sus propiedades antiinflamatorias y antioxidantes. El presente trabajo pretende profundizar en el mecanismo molecular responsable del efecto neuroprotector de estas terapias. En concreto se centra en la modulación de la proteína SIRT1 debido, entre otras cosas, a su papel regulador de la vía de señalización proinflamatoria NF-kB. Los resultados muestran una reducción en los niveles de SIRT1 en el hipocampo de rata durante el envejecimiento, que es contrarrestado tanto por el ejercicio físico como por la acción combinada del ejercicio y la administración exógena del antioxidante catequina. Estos efectos se correlacionan con una mejora en las capacidades cognitivas durante el envejecimiento producido por ambos tratamientos. Sorprendentemente, no se observa ningún efecto sumatorio por ambos tratamientos.
[eng] In recent decades there has been an increase in life expectancy, which has supposed an increase in the incidence of diseases associated with aging. The theory of oxidative stress is one of the most accepted theories to explain the causes of aging, as well as the increase in the inflammatory response in which oxidative stress could be involved. The chronic inflammatory phenotype characteristic of aging contributes to the decline of physiological functions, which is one of the main risk factors for different pathologies such as neurodegenerative diseases. Nowadays, aging is an object of research to determine molecular bases that produce it and modulate it, in order to prevent or delay their consequences. Among the therapies we could highlight the performance of physical exercise and administration of antioxidants for their anti – inflammatory and antioxidant properties. The present work pretends to go into detail about the responsible molecular mechanism for the neuroprotective effect of these therapies. Specifically, it focuses on the modulation of the SIRT1 protein due to its regulation role in the NF – kB proinflammatory signaling pathway, between other things. The results show a reduction in the SIRT1 levels in the rat hippocampus during aging, which is counteracted by physical exercise and the combined action of exercise and the administration of the catechin antioxidant. These effects correlate with an improvement in cognitive abilities during aging produced by both treatments. Surprisingly, no added effect is observed for both treatments.
