[eng] Colorectal cancer is the third most common cancer worldwide. The development of colorectal
cancer is influenced by environmental factors, genetic factors and inflammatory conditions.
Inflammation is a physiological response mechanism against injurious stimuli constituting a key
component in the microenvironment that determines tumoral induction, progression and
invasiveness. Cytokines are the main mediators in establishing this microenvironment through
cell-cell signalling and gen expression modulation in cells that conform it. In this work the proteic
expression levels of the transcription factors NF-κB and STAT3, the regulator IκB, as well as the
COX-2 protein in the stages II, III and IV of colorectal tumor samples have been studied through
Western blot analysis in order to determine the way in which the inflammation influences
colorectal cancer tumorigenesis.The results show that the expression levels of NF-κB and
STAT3 tend to increase and that COX-2 expression decreases in stage IV due to a greater
inhibition of NF-κB by IκB, suggesting that STAT3 may also be inhibited at a cytosolic level.
These conclusions could set the basis for new customized therapeutic strategies design based
on the stage of development of each tumor.
[cat] El càncer colorectal és el tercer tipus de càncer més comú a nivell mundial. El
desenvolupament de càncer colorectal es veu influenciat per factors ambientals, factors
genètics i condicions inflamatòries. La inflamació és un mecanisme fisiològic de resposta
enfront estimuls lesius que constitueix un component clau en l’establiment del microambient
que determina la inducció, progressió i invasió tumoral. Les citoquines són els mediadors
principals en l’establiment d’aquest microambient mitjançant senyalització intracel·lular i
modulació de l’expressió gènica de les cèl·lules que el conformen. En aquest treball s’han
estudiat els nivells d’expressió proteica dels factors de transcripció NF-κB i STAT3, el regulador
IκB, així com la proteïna COX-2 en els estadis II, III i IV de mostres de tumor colorectal
mitjançant Western blot. L’objectiu és determinar la manera per la que la inflamació influeix en la
tumorigènesi del cancer colorectal. Els resultats mostren que els nivells d’expressió de NF-κB i
STAT3 tendeixen a augmentar i que l’expressió de COX-2 disminueix en l’estadi IV a causa
d’una major inhibició de NF-κB per part de IκB, suggerint que STAT3 també pot estar inhibit a
nivell citosòlic. Aquestes conclusions podrien establir la base pel disseny de noves estratègies
terapèutiques personalitzades en funció de l’estadi de desenvolupament de cada tumor.
