Identificació molecular dels crisomèlids de l'illa de sa Dragonera (Mallorca) i de les seves plantes hostes

[cat] Els crisomèlids són un grup molt diversificat d’escarabats que es poden trobar pràcticament a tots els ecosistemes terrestres del món. La seva abundància juntament amb els seus hàbits herbívors els converteixen en uns components rellevants a les xarxes tròfiques. Per aquest motiu, la correcta identificació de les espècies de crisomèlids i de les seves interaccions tròfiques pot resultar important de cara a conèixer els detalls del funcionament dels ecosistemes on viuen. Al nostre arxipèlag Balear es distribueixen per totes les illes, però en aquest treball ens centrem en els de l’illa de Sa Dragonera (Mallorca), on fins ara s’han identificat 15 espècies classificades en 3 subfamílies (Alticinae, Clytrinae i Cryptocephalinae). L’objectiu del present treball és du a terme un estudi centrat en 5 d’aquestes espècies amb la finalitat de determinar des d'un punt de vista molecular l’espècie a la que pertanyen, així com l’alimentació de cada una d’elles. Es va extreure l’ADN a partir del cos complet dels insectes, de manera que es va facilitar l’obtenció de l’ADN dels propis escarabats i també l’ADN de les plantes hostes del seu interior. Les identificacions de les diferents espècies es van realitzar mitjançant la tècnica dels codis de barres d'ADN o DNA barcoding, amb la seqüenciació del gen mitocondrial cox1, i dels gens cloroplàstics psbA-trnH, rbcL i trnL. Les seqüències obtingudes es van contrastar amb les bases de dades GenBank i BOLD per a la seva identificació taxonòmica mitjançant comparació per similaritat i inferència filogenètica. Els resultats obtinguts han permet la identificació molecular de les espècies de crisomèlids estudiades així com les plantes que havien ingerit. Les espècies dieta que hem trobat són consistents amb la informació prèviament disponible a la bibliografia, sent principalment del gènere Pistacia, Erodium i la família Asteraceae les més comuns.

[eng] Chrysomelids are a highly diversified group of beetles that can be found almost in all of terrestrial ecosystems. Their abundance along with their herbivorous habits make them a relevant component in the trophic networks. For this reason, the correct identification of chrysomelid species and their trophic interactions can be important in order to know the details of the functioning of the ecosystems in which they live. In our Balearic archipelago, they are distributed throughout all the main islands, but in this work we focus on the island of Sa Dragonera (Mallorca), where up to now, 15 species have been identified in 3 subfamilies (Alticinae, Clytrinae and Cryptocephalinae). The objective of this work is to carry out a study focused on 5 of such species in order to determine, from a molecular point of view, to which species they belong, as well as their diet habits. The DNA was extracted from the complete body of the insects, thus allowing the retrieval of the DNA from the beetles themselves and also the DNA from the host plants inside them. The identification of the different species were performed using the DNA barcoding technique, with sequencing of the mitochondrial cox1 gene, and chloroplastic genes psbA-trnH, rbcL and trnL. The obtained sequences were contrasted with the GenBank and BOLD databases for their taxonomic identification by similarity and also by phylogenetic inference. The results obtained allowed the molecular identification chrysomelid species as well as the plants that they had ingested. The host plant species we have found are consistent with the available information in the literature, including the genera Pistacia, Erodium and the Asteraceae plant family.
