[cat] Pseudomonas aeruginosa és considerat un dels patògens oportunistes més
importants, causant d’una àmplia diversitat d’infeccions a l’ésser humà. Destaca la
seva importància clínica a les infeccions respiratòries cròniques com la fibrosi quística,
la bronquièctasi i la malaltia pulmonar obstructiva crònica (MPOC). El patogen es
caracteritza per presentar una gran capacitat de desenvolupar resistències als
antibiòtics, ja sigui per mutacions cromosòmiques o per adquisició gènica. En aquest
estudi, hem avaluat l’eficàcia terapèutica i la dinàmica de resistència antibiòtica
damunt els biofilms de P. aeruginosa tractats amb un nou antibiòtic (NA1). Es varen
utilitzar les soques de laboratori PAO1 i PAOMA (mucoide), a més de la soca clínica
146-HSE (soca epidèmica de Liverpool[LES-1]). Inicialment, es van determinar les CMI
de l’antibiòtic per aquestes soques a través del mètode de dilució en brou amb la
intenció d’establir les concentracions antibiòtiques adequades pel tractament dels
biofilms. Els biofilms es varen desenvolupar fent ús del model de tractament en
cambra de flux. Les concentracions de NA1 escollides per dur a terme el tractament
dels biofilms foren 0,5 µg/ml, 2 µg/ml i 8 µg/ml, equivalents a 1x, 4x i 16x la CMI de la
soca PAO1, respectivament. Es va determinar el nombre de cèl·lules viables (UFC) i
de mutants resistents. Per altra banda, la dinàmica estructural del biofilm es va poder
seguir mitjançant microscòpia confocal. El processament de les imatges obtingudes es
va realitzar amb els programes de software IMARIS i COMSTAT. L’ús de les
concentracions de 0,5 µg/ml o 2 µg/ml de NA1 va contribuir a la selecció de mutants
resistents a NA1. No obstant, l’ús d’una concentració de 8 μg/ml va reduir de manera
eficient el nombre de cèl·lules viables als biofilms de P. aeruginosa, a més de prevenir
la selecció de mutants resistents. Aquests resultats suggereixen que NA1 podria
mostrar bona activitat enfront els biofilms de P. aeruginosa, podent arribar a
considerar-se en un futur una nova i eficaç alternativa terapèutica. |

[eng] Pseudomonas aeruginosa is considered one of the most important opportunistic
pathogens, cause of a wide variety of infections in human beings. Highlights its clinical
significance in chronic respiratory infections such as Cystic Fibrosis, bronchiectasis
and chronic obstructive pulmonary disease (COPD). The pathogen is characterized by
presenting a great ability to develop resistance to antibiotic, either for chromosomal
mutations or gene acquisition. In this study, we have evaluated the therapeutic efficacy
and dynamics of antibiotic resistance in P. aeruginosa biofilms treated with a new
antibiotic (NA1). Laboratory strains PAO1 and PAOMA (mucoid), in addition to 146-
HSE (Liverpool epidemic strain [LES-1]), were used. Initially, we determined the
antibiotic MIC by these strains through broth dilution method, with the intention of
establishing adequate antibiotic concentrations for the treatment of biofilms. Biofilms
were developed using the treatment model in flow cell. NA1 concentrations chosen to
carry out the biofilm treatment were 0,5 µg/ml, 2 µg/ml and 8 µg/ml, equivalent to 1x, 4x
and 16x MIC of strain PAO1, respectively. The number of viable cell (CFU) and
resistant mutants was determined. On the other hand, biofilm structural dynamics could
be followed by confocal microscopy. The processing of the images obtained was
carried out using IMARIS and COMSTAT software programs. The use of
concentrations of 0,5 µg/ml or 2 µg/ml of NA1 contributed to the selection of resistant
mutants to NA1. However, the use of a concentration of 8 µg/ml reduced efficiently the
number of viable cells in P. aeruginosa biofilms, as well as preventing the selection of
resistant mutants. These results suggest that NA1 could show a good activity against
P. aeruginosa biofilms and could be considered a new and effective therapeutic
alternative in the future |
