[cat] Les micorrizes són associacions simbiòtiques entre determinats fongs del sòl i les arrels de les plantes. Els fongs formadors de micorrizes aporten un gran nombre de beneficis a la planta hoste, ja que augmenten la superfície d'absorció de la planta facilitant l'absorció de nutrients i aigua. A més, també augmenten la tolerància de la planta hoste a l'atac de patògens, i protegeixen contra el canvi climàtic. Concretament en la vinya s'ha demostrat que la formació de micorrizes afavoreix l'acumulació de composts antioxidants propis del raïm, augmentant l'activitat antioxidant de la vinya. Donada la importància de les micorrizes en els cultius agrícoles, una correcta identificació de les espècies de fongs formadores de micorrizes és important per garantir l'èxit dels cultius. Els primers estudis taxonòmics i sistemàtics foren duts a terme per Tuslane (1845) i Thaxer (1922), que identificaren que el major nombre de fongs formadors de micorrizes pertanyien a la Divisió Glomeromycota. L'estudi morfològic ha anat acompanyat de l'ús de característiques bioquímiques permetent la separació dels taxons en distints nivells. La identificació a partir de les característiques morfològiques de les espores té una sèrie de limitacions, com el fet de distingir entre espores viables i les no viables. Per aquest motiu, l'aplicació de tècniques moleculars per a la correcta identificació i classificació és important de cara a conèixer més a fons aquest grup de fongs. En el present estudi s'ha extret ADN de 32 mostres d'arrels de vinya amb l'objectiu d'establir si són susceptibles a ser amplificades i seqüenciades amb marcadors moleculars específics de fongs formadors de micorrizes. De les identificacions realitzades, aquelles relatives al gènere Glomus es corresponen amb fongs formadors de micorrizes. Per a la majoria de mostres es va poder arribar al nivell de gènere del fong al qual pertanyien les seqüències obtingudes i, en menor mesura, al nivell d'espècie. |

[eng] Mycorrhizae are symbiotic associations between some soil fungi and plant roots. Mycorrhizal fungi provide a large number of benefits to the host plant by increasing the absorption surface of the plant, facilitating the absorption of nutrients and water. In addition, they also increase the host plant's tolerance to pathogen attack and protect against climate change. Specifically, in the vineyard it has been demonstrated that the formation of mycorrhizae stimulates the accumulation of antioxidant compounds typical of the grape, increasing the antioxidant activity of the vineyard. Given the importance of mycorrhizae in agricultural crops, a correct identification of the species of fungi that form mycorrhizae is essential to ensure crop success. The first taxonomic and systematic studies were carried out by Tuslane (1845) and Thaxer (1922), who identified that the largest number of mycorrhizal fungi belonged to the Glomeromycota Division. The morphological study has been accompanied using biochemical characteristics allowing the separation of taxa at different levels. Identification from the morphological characteristics of spores has several limitations, such as having to distinguish between viable and non-viable spores. For this reason, the implementation of molecular approaches for the correct identification and classification is important in order to know more about this group of fungi. Here, we extract DNA from 32 vine root samples in order to establish whether they are susceptible to amplification and sequencing using mycorrhizal fungi specific molecular markers. Of the sequences obtained, those relating to the genus Glomus correspond to mycorrhizal fungi. For most samples it was possible to reach the genus level of the fungus to which the sequences obtained belong and, to a lesser extent, to the species level. |
