[cat] La present Tesi Doctoral té com a objectiu portar a terme estudis que formen part del procés de desenvolupament preclínic del myo-inositol hexafosfat com a fàrmac per al tractament de malalties relacionades amb processos patològics de calcificació. Concretament, s’han realitzat estudis de farmacocinètica, prèvia qualificació i validació dels corresponents mètodes analítics en matrius biològiques.
La disponibilitat d’eines adequades per a la determinació del perfil d’impureses i la potència del principi actiu utilitzat pels diversos estudis en el procés de desenvolupament d’un fàrmac resulta una primera etapa crítica que requereix el desenvolupament i validació dels mètodes bioanalítics adequats d’acord amb les directrius marcades per la ICH (International Conference on Harmonisation).
Aquests treballs analítics representen l’eina fonamental per tal d’estudiar posteriorment el comportament farmacocinètic del myo-inositol hexafosfat. La informació que se n’obtingui, completarà la dels estudis preclínics, i permetran obtenir la dosi terapèutica estimada per tal d’iniciar els estudis clínics.
[eng] The aim of this Doctoral Thesis is to develop some of the required studies related to the preclinical process for the approvement of myo-inositol hexaphosphate as a drug for the treatment of diseases associated with pathological calcifications. In particular, qualification and validation of analytical methods in biological matrices has been performed for the determination of the pharmacokinetic profile of the product.
The availability of suitable tools to determine the impurities profile and the capability of the API used for several studies in the process of drug development is a critical step that requires the development and validation of bioanalytical methods following ICH guidelines (International Conference on Harmonization).
This analytical work represents the fundamental tool for the study of the pharmacokinetic profile of myo-inositol hexaphosphate. The obtained data will complete the preclinical studies of safety and will allow the determination of the estimated therapeutic dose to be used in Clinical Phase.
