[cat] El sistema circadià permet la coordinació de les funcions vitals de l’organisme en relació amb l’entorn i es basa en l’expressió d’una sèrie de gens que comporten un rellotge molecular, permetent que les cèl·lules puguin actuar de manera independent. Aquests gens són expressats a regions del cervell com ara l’hipocamp, regulant les seves funcions principals: l’aprenentatge i la memòria. Estudis previs han comprovat que aquestes funcions es troben íntimament relacionades amb la neurogènesi adulta, que consisteix en la generació de noves neurones a partir de cèl·lules progenitores. En aquest treball s’ha estudiat la regulació circadiana de la neurogènesi hipocampal en rates mascle adultes Sprague-Dawley, les quals han estat sacrificades en unes hores determinades, que corresponen a la fase de llum (8 h-20 h) o a la fase d’obscuritat (20 h-8 h). A partir d’una immunohistoquímica basada en l’ús del marcador endogen Ki-67, s’ha observat que la proliferació cel·lular és significativament superior durant la fase de foscor. Així doncs, aquest procés és major durant la nit subjectiva, que correspon al període actiu de les rates adultes. Aquests resultats, juntament amb altres estudis realitzats en models animals knock-out pels gens del rellotge molecular i sobre alguns factors reguladors de la neurogènesi, han permès obrir l’horitzó a l’estudi d’aspectes relacionats amb l’aprenentatge i la memòria, a més de conèixer de manera més profunda algunes malalties tant importants en l’actualitat com l’Alzheimer.
[eng] The circadian system allows the coordination of the organism's vital functions in relation to the environment. This system is based on the expression of a series of genes that involve a molecular clock, allowing cells to act independently. These genes are expressed in some regions of the brain such as hippocampus, regulating their main functions: learning and memory. Previous studies have shown that these functions are closely related with adult neurogenesis, which consists of the generation of new neurons from progenitor cells. In this report, the circadian regulation of hippocampal neurogenesis in adult male Sprague-Dawley rats has been studied. Rats were sacrificed within a given time, corresponding to the light phase (8 h–20 h) or the dark phase (20 h-8 h). Via the realization of an immunohistochemistry based on the use of the Ki-67 endogen marker, cellular proliferation was observed to be significantly higher during the dark phase. Thus, this process is greater during the subjective night, corresponding to the active period of adult rats. These results, along with other studies conducted on knock-out animal models for molecular clock genes and some regulatory factors of neurogenesis, have allowed the opening of new horizons in the study of learning and memory-related aspects, as well as increasing the knowledge about some important neural diseases such as Alzheimer.
