[spa] La leishmaniosis una de las enfermedades más extendidas entre las comunidades animales, está causada por la transmisión de Leishmania infantum, a través de mosquitos del género Phlebotomus. El método típico de detección de este parásito es mediante la técnica de la PCR cuantitativa (qPCR). En este estudio, se propone un método alternativo de detección del parásito mediante la utilización de la técnica de droplet digital PCR (ddPCR), más sensible, en muestras de sangre periférica y cerumen de oído. Como tratamiento alternativo para la leishmaniosis, se ha propuesto complementar el tratamiento estándar de Alopurinol y Glucantime con células madre mesenquimales (MSC). El estudio se compone de dos grupos de animales, uno de ellos tratado con el tratamiento estándar y el otro con el complemento de células madre mesenquimales. Muestras de sangre y cerumen se extrajeron en los distintos tiempos de estudio. A partir de las extracciones de ADN de estas muestras se realizó la qPCR y la ddPCR, se diseñaron primers y sondas para la detección del parásito. A demás se realizó el análisis de diversos parámetros bioquímicos. Las diferencias entre grupos se determinaron mediante ANOVAS, para distintos parámetros, cómo en ciclo qPCR o la concentración de creatinina, entre otros. No se observaron diferencias estadísticas entre grupos, pese a ello se pueden observar pequeñas tendencias en la reducción de parámetros, cómo la concentración de creatinina o la de proteínas en orina, relacionados con el daño renal de los animales.
[eng] Leishmaniasis, one of the most widespread diseases among the animal community, is caused by the transmission of Leishmania infantum, through mosquitoes of the genus Phlebotomus. The typical method of detection of this parasite is by using the quantitative PCR technique (qPCR). In this study, an alternative method of detecting the parasite is proposed using the more sensitive droplet digital PCR (ddPCR) technique in peripheral blood and ear wax samples. As an alternative treatment for leishmaniasis, it has been proposed to complement the standard treatment of Allopurinol and Glucantime with mesenchymal stem cells (MSC). The study consists of two groups of animals, one of them treated with the standard treatment and the other with the complement of mesenchymal stem cells. Blood and cerumen samples were obtained at the different study times. From the DNA extractions of these samples, qPCR and ddPCR were performed, primers and probes were designed for the detection of the parasite. In addition, the analysis of various biochemical parameters was performed. The differences between groups were determined using ANOVAS, for different parameters, such as qPCR cycle or creatinine concentration, among others. No statistical differences were observed between groups, despite this, small trends can be observed in the reduction of parameters, such as the concentration of creatinine or that of proteins in urine, related to kidney damage in the animals.
