[cat] L’actual pandèmia de la COVID-19, causada pel SARS-CoV-2 ha fet posar el focus a les autoritats sanitàries i a la comunitat científica sobre la transmissió d’aquest coronavirus. Una de les fonts de transmissibilitat del virus que s’ha estat estudiant és a través de microgotes d’origen biològic de llarga duració dins l’ambient, anomenats bioaerosols. Davant aquest fet, aquest treball proposa millorar alguns aspectes d’una metodologia que permeti quantificar els nivells de SARS-CoV-2 dels bioaerosols, la qual cosa és necessària per estimar el risc d’infecció i facilitar la gestió de desinfecció. Per tal de trobar una resposta al problema que es planteja, s’han dut a terme diferents experiments en els que s’analitza la influència del número de persones presents en un espai tancat, la ventilació, el volum d’aire recollit i, el medi de recol·lecció en la quantificació d’ARN a bioaerosols, i l’estudi dels encebadors més òptims per a la detecció del genoma de SARS-CoV-2. Com a resultats més rellevants destaquem que l’ARN aïllat de les mostres d’aire no prové exclusivament dels bioaerosols de la respiració sinó que hi ha un alt percentatge que prové de la microbiota natural de l’aire. S’ha pogut establir una correlació positiva entre el número de persones i el CO2 que hi ha a un espai tancat, però aquests dos paràmetres presenten una correlació dèbil amb l’ARN aïllat. També s’ha pogut comprovar que les recol·leccions de volums d’aire més grans no suposen un increment en la quantitat d’ARN recollit. El medi de recol·lecció hidròfob ha permès recollir més ARN de l’aire. Finalment, s’han establert tres encebadors N i N1 (reconeixen el gen de la nucleocàpsida) i la seqüència ORF1ab del genoma de SARS-CoV-2 que podrien ser més òptims per a la detecció del virus a bioaerosols. L’encebador que reconeix la seqüència ORF1ab destaca per tenir una major eficiència. L’encebador que reconeix la seqüència N és el més específic en la detecció del virus. Finalment, l’encebador que reconeix el gen N1 destaca per tenir una major sensibilitat en la detecció de SARS-CoV-2. L’estudi dels factors metodològics que aquí s’han analitzat contribuirà a millorar el protocol de detecció de SARS-CoV-2 a bioaerosols.
[eng] The current COVID
19 pandemic, caused by SARS CoV 2, has put the health authorities and
the scientific community in the spotlight on the transmissi on of this coronavirus. One of the
sources of transmissibility of the virus that has been studied is through microdroplets of long
biological origin that stays more time in the air , called Given this fact, this work
proposes to improve some aspe cts of a methodology to quantify the SARS CoV 2 levels of
bioaerosols, which is necessary to estimate the risk of infection and facilitate disinfection
management. To find an answer to the problem posed, different experiments have been carried
out in which the influence of the number of people present in an enclosed space, ventilation,
the volume of air collected and the collection media on the quantification of RNA to bioaerosols in analysed, as well as
the study of the most optimal primers for the detecti on of the genome
of SARS CoV 2. As more relevant results, we note that the RNA isolated from air samples
does not come exclusively from the bioaerosols of respiration but there is a high percentage
that comes from the natural microbiota of the air. A posit ive correlation has been established
between the number of people and the CO 2 levels in an enclosed space, but these two
parameters have a weak correlation with the amount of isolated RNA. It has also been shown
that collecting larger volumes of air does n ot increase the amount of RNA collected. The
hydrophobic collection medium has made it possible to collect more RNA from the air. Finally,
three primers N and N1 (recognizing the nucleocapsid gene) and the ORF1ab sequence of the
SARS CoV 2 genome have been established that may be a better option for viru s detection.
The primer s that recognize the ORF1ab sequence stands out for its greater efficiency. The
primer s that recognize the N sequence is the most specific in detecting the virus. Finally, the
primer s that recognize the N1 gene stands out for having a higher sensitivity in the detection
of SARS CoV 2. The methodological factors studied here will contribute t o improving the
SARS CoV 2 detection protocol in bioaerosols.
