[spa] La Atrofia Muscular Espinal es una enfermedad genética (rara) que afecta a 1/11.000 nacidos vivos. Puede tener una severidad muy variable, pero los fenotipos más graves son las AMEs de los tipos 0 y 1, que tienen una corta esperanza de vida. Este trastorno no tenía tratamiento farmacológico ninguno hasta el 2016, año en el cual se aprobó el Nusinersen (oligonucleótido antisentido) que es un medicamento que se administra de manera intatecal. Esta revisión bibliográfica tiene como propósito analizar si el medicamento Nusinersen es eficaz y seguro para este tipo de pacientes. Se han buscado artículos en 3 bases de datos diferentes (PubMed, CINAHL, Cochrane), acotando la búsqueda 10 años (2011-2021) y eligiendo artículos solo en inglés (e incluso, alguno en español). La mayor parte de artículos encontrados fueron revisiones sistemáticas de ensayos clínicos y de casos y controles. De los 80 artículos que cumplían el tercer nivel de búsqueda, fueron incluidos en este trabajo 23 que cumplían los criterios de inclusión y exclusión.
[eng] Spinal Muscular Atrophy is a genetic disease (rare) affecting 1/11,000 live births. It can have a very variable severity, but the most severe phenotypes are SMA types 0 and 1, which have a short life expectancy. This disorder had no pharmacological treatment until 2016, when Nusinersen (antisense oligonucleotide) was approved, which is a drug that is administered intrathecally.
This literature review aims to analyze whether the drug Nusinersen is effective and safe for this type of patients. Articles were searched in 3 different databases (PubMed, CINAHL, Cochrane), limiting the search to 10 years (2011-2021) and choosing articles only in English (and even some in Spanish). Most of the articles found were systematic reviews of clinical trials and case-control studies. Of the 80 articles that met the third level of the search, 23 that met the inclusion and exclusion criteria were included in this study.
