[cat]L’envelliment és un procés progressiu i universal que provoca el declivi de la funcionalitat de
l’organisme, augmentant el risc de mort. Es caracteritza per un increment del dany oxidatiu degut a
la producció elevada de radicals lliures i espècies reactives d’oxigen (ROS), lo que potencia el risc de
patir malalties degeneratives. El cervell és l’òrgan que presenta la major taxa de consum d’oxigen, de
forma que és més susceptible al dany oxidatiu i al deteriorament de les capacitats cognitives i
motores. Els ROS actuen sobre els enzims implicats en la síntesi de les monoamines, reduint la seva
activitat en la síntesi de neurotransmissors, com la dopamina, la serotonina i la noradrenalina, en les
regions cerebrals principalment implicades en els processos cognitius i motors: l’hipocamp i l’estriat.
S’ha demostrat que l’administració farmacològica de substàncies antioxidants, com polifenols,
resveratrol, melatonina, vitamina E..., presents de forma natural en diversos aliments, millora les
funcions cerebrals incrementant els nivells d’aquestes monoamines. Alguns tenen la capacitat
d’activar les sirtuïnes, especialment la SIRT1, la qual s’ha relacionat amb els efectes beneficiosos de
la restricció calòrica i l’augment de l’esperança de vida. Aquesta activa diverses vies fisiològiques
implicades en evitar l’envelliment i presenta una acció beneficiosa sobre el cervell. En aquest treball
es pretenia estudiar els efectes d’una dieta rica en antioxidants sobre l’envelliment cerebral de rates
velles mitjançant la realització de diverses proves comportamentals, la determinació dels nivells de
monoamines i la quantificació de la SIRT1 en el cervell. Les rates tractades demostraren un cert
increment en les monoamines i la SIRT1, però les funcions cognitives i motores no milloraren de
manera significativa. Per tant, les quantitats d’antioxidants presents en els aliments no serien
suficients per retardar l’envelliment del cervell i es necessitaria una major aportació per aconseguir
els resultats esperats. |

[eng]Aging is a universal process that causes progressive decline of an organism’s function, increasing the
risk of death. It is characterized by an increased oxidative damage due to the high production of free
radicals and reactive oxygen species (ROS), which increases the risk of degenerative diseases. The
brain is the organ with the highest rate of oxygen consumption, so it is more susceptible to oxidative
damage and impaired motor and cognitive skills. ROS act on enzymes involved in the synthesis of
monoamines, reducing its activity in the synthesis of neurotransmitters, such as dopamine, serotonin
and norepinephrine, in the brain regions mainly involved in cognitive and motor abilities: the
hippocampus and the striatum. It has been shown that a pharmacological administration of
antioxidants, such as polyphenols, resveratrol, melatonin, vitamin E..., present naturally in various
foods, improves brain function by increasing the levels of these monoamines. Some of them have the
ability to activate sirtuins, particularly SIRT1, which is related to the beneficial effects of caloric
restriction and increased lifespan. SIRT1 activates different physiological pathways involved in
avoiding the effects of aging and has a beneficial effect on the brain. The aim of this work was to
study the effects of a diet rich in antioxidants on the brain aging of old rats by performing behavioral
tests, the determination of monoamines levels and the quantification of SIRT1 in the brain. Treated
rats showed an increase in monoamines and SIRT1, but motor and cognitive functions did not
improve significantly. Therefore, the amount of antioxidants present in food wouldn’t be enough to
slow down brain aging and it would take a major contribution to achieve the expected results. |
