[spa]La matriz ósea desmineralizada (DBM) es un aloinjerto óseo con propiedades
osteoinductoras muy utilizado para promover la formación de nuevo hueso. Este
potencial osteoinductivo es debido a las proteínas morfogenéticas óseas (BMP) que
contiene, y a otras moléculas como miembros de la familia TGF-β o la metaloproteasa
de la matriz MMP2. La DBM se obtiene a partir del hueso cortical del fémur de
donantes, tras la molienda, lavado y desmineralización. Para su aplicación, es
necesario formular la DBM con un excipiente que, además de facilitar su manipulación
e implementación, potencie su actividad osteoinductora. En este estudio, se han
evaluado tres excipientes (glicerol, ácido hialurónico y metacriloilo de gelatina
(GelMA)) para formular la DBM y se ha investigado su efecto sobre la diferenciación
osteogénica de la línea de células musculares C2C12. Todas las muestras de DBM de
los donantes analizados indujeron la actividad ALP en células musculares, indicativo de
la diferenciación osteogénica. Además, los extractos proteicos de las DBM analizados
mostraron niveles cuantificables de BMP2. De los excipientes investigados, ninguno
resultó tóxico para las células C2C12 y el GelMA fue el excipiente que mostró una
mayor inducción de la actividad ALP en estas células, cuando se formularon a una
concentración de DBM al 30%. Esta concentración es la que mostró una mejor
consistencia y maleabilidad. En conclusión, el GelMA es un excipiente que podría ser
usado para la formulación de la DBM y su aplicación clínica en humanos
[eng]The demineralized bone matrix (DBM) is a bone allograft with osteoinductive
properties widely used to promote the formation of new bone. This osteoinductive
potential is due to the presence of bone morphogenetic proteins (BMP) and other
proteins like members of the TGF-β family or the matrix metalloproteinase MMP2.
DBM is obtained from human femoral cortical bone, after grinding, washing and
demineralization. For its application, it is necessary to formulate the DBM with an
excipient that, in addition to facilitating handling and implementation, it also enhances
its osteoinductive activity. In this study, we evaluated three excipients (glycerol,
hyaluronic acid and gelatin methacryloyl (GelMA)) to formulate the DBM and we have
investigated its effect on osteogenic differentiation of the C2C12 muscle cell line. All of
the DBM donor samples that were analyzed induced the ALP activity in muscle cells,
indicative of osteogenic differentiation. In addition, the protein extracts of DBM
showed measurable levels of BMP2. Of the excipients tested, none were toxic to
C2C12 cells and the GelMA was the excipient that showed a greater induction of ALP
activity in these cells when formulated at a concentration of 30% DBM. This
concentration was the one that showed better consistency and malleability. In
conclusion, the GelMA is an excipient that could be used to formulate the DBM for its
clinical application in humans
