[cat] Els biomarcadors permeten dur a terme estudis de biomonitorització de la contaminació marina, en aquest cas, a Mytilus galloprovincialis per dur a terme un estudi comparatiu entre tres zones de Sicília, Priolo, on hi ha una refineria, i dues zones netes, Brucoli i Messina. L’estudi ha permès determinar l’estat oxidatiu dels musclos mitjançant tota una sèrie de biomarcadors, així com, el dany oxidatiu ocasionat com a conseqüència de l’augment de la producció de les espècies reactives d’oxigen (ROS) generades pel sistema de detoxificació. S’ha dut a terme la determinació de l‘activitat enzimàtica de la catalasa (CAT), com enzim constituent del sistema antioxidant, i de la glutatió-S-transferasa (GST), com a component del sistema de detoxificació de xenobiòtics de fase II, mitjançant tècniques espectrofotomètriques. Per altra banda, s’ha realitzat l’estudi de la citocrom P4501A, constituent del sistema de detoxificació de fase I, mitjançat la determinació de la seva expressió amb la realització d’un PCR a temps real (PCR-RT). Juntament amb aquest, s’ha dut a terme la determinació de l’expressió dels gens de la CAT i la GST, així com de les metal·lotioneïnes (MT10 i MT20) per observar la seva inducció per metalls pesants. Finalment, s’ha estudiat el dany oxidatiu observant els canvis en la peroxidació lipídica, determinant la concentració de malondialdehid (MDA) i, per altra banda, determinant l’activitat de l’acetilcolinesterasa (AChE), ambdues mitjançant tècniques espectrofotomètriques. Els resultats obtinguts mostren com els musclos de la zona contaminada (Priolo) presenten un major estrès oxidatiu, mostrant una elevació dels enzims del sistema de defensa antioxidant i de detoxificació, així mateix, també han mostrat una elevació del nivell de MTs i dels indicadors de dany oxidatiu, fet que indica que l’augment del sistema de defensa antioxidant no ha estat suficient per fer front a la producció dels ROS.
[eng] Biomarkers are able to realize marine pollution biomonitoring assays, in this case, in Mytilus galloprovincialis to perform a comparative study of tree locations in Sicily, Priolo, where there is a refinery, and two clean zones, Brucoli and Messina. This study has enabled to determinate the oxidative status of the mussels, using a group of biomarkers, and the oxidative damage produced as a consequence of the production increase of the reactive oxygen species (ROS) that are generated by the detoxification system. It has been performed the enzymatic activity determination of catalase (CAT), as an antioxidant system constituent, and glutathione-S-transferase (GST), as a phase II detoxification system constituent, by means of spectrophotometric methods. Otherwise, it has been performed a gene expression determination of cytochrome P4501A, as a constituent of the phase I detoxification system using a real time PCR (RT-PCR). Furthermore, it has been determined the gene expression of CAT and GST, and metallothioneins (MT10 and MT20) in order to observe the metal induction of the last ones. Finally, it has been studied the oxidative damage, concretely the lipid peroxidation variation, determining the malondialdehyde (MDA) concentration, and the acetylcholinesterase (AChE) activity variation, both by spectrophotometric techniques. Results obtained shown an increase of the oxidative stress in the mussels of the polluted location (Priolo), with an increase of the defense and detoxification enzymes levels, MTs levels and in the oxidative damage indicators that showed that the rise in antioxidant capacity to remove the excess production of ROS was not enough to avoid oxidative damage
