Contribució europea a l’estructura genètica de Valles Calchaquíes (Nord-oest d’Argentina)

Abstract

[cat] El DNA mitocondrial presenta unes característiques úniques que expliquen la seva utilitat en Genètica Forense i de Poblacions. Conseqüentment, en aquest treball s’ha estudiat la regió control del DNA mitocondrial de 25 individus procedents de les Valles Calchaquíes. L’estudi d’aquestes poblacions és especialment interessant ja que constitueixen nuclis rurals relativament aïllats, amb una baixa densitat de població i poca migració. A més, la conquesta espanyola ha suposat un esdeveniment històric rellevant en l’estructura genètica dels seus habitants, els quals són resultat de la mescla d’europeus, amerindis i africans. L’objectiu de l’estudi és estimar la proporció de mescla que presenten per via materna els habitants de les poblacions de les Valles Calchaquíes. Per aquest motiu, s’ha portat a terme l’extracció a partir de sang seca, l’amplificació i la seqüenciació de la regió D-loop del mtDNA; obtenint-se com a resultat els haplogrups associats al genoma mitocondrial de 21 individus. S’ha determinat que totes les mostres presenten haplogrups amerindis, essent l’A i el B els més abundants (33’33%), seguit del D (19’05%) i finament el C (14’29%). La manca de contribució europea i africana per línia materna en les mostres estudiades, possiblement és deguda a l’aïllament d’aquestes poblacions, tot i que un estudi amb més mostres seria necessari per a obtenir resultats estadísticament significatius. Les recerques bibliogràfiques indiquen que mitjançant l’anàlisi de marcadors uniparentals i biparentals del genoma s’estimen diferents graus de mescla, essent especialment interessants els obtinguts en estudis on s’analitzen els polimorfismes del cromosoma Y, on s’indica que el component amerindi és el minoritari. Aquest biaix lligat al sexe possiblement és conseqüència de la direccionalitat dels matrimonis entre homes europeus i dones ameríndies.

[eng] Mitochondrial DNA presents unique characteristics that explain its utility in Forensic and Populations Genetics. For this reason, in this study, the mitochondrial DNA control region of 25 individuals from the Calchaquí Valleys has been studied. The analysis of these populations is especially interesting because they live in small isolated groups, with low density and migration rates. Moreover, the Spanish conquest has been an important historic event for the genetic structure of its habitants, which are a mix of European, Amerindian and Africans. The purpose of this study is to estimate the admixture ratio that the inhabitants of the Calchaquí Valleys present through maternal lineages. For this reason the extraction of DNA from dry blood, the amplification and the sequencing of the D-loop region has been carried out. The haplogroups of 21 individuals has been obtained as a result. All of these samples have Amerindian haplogroups, being the A and the B the most frequent (33’33%), followed by the D (19’05%) and finally the C (14’29%). The absence of European and African contribution in maternal lineages in the studied samples, possibly is caused by the isolation of these populations. However, another study with more samples is required to obtain statistically significant results. Bibliographic searches indicate that the analysis of uniparental and biparental markers distributed throughout the genome estimate different admixture ratios. The results obtained on the studies where the Y chromosome polymorphisms are analysed are especially interesting, indicating that the Amerindian is the minor component. This sex-biased contribution is likely due to directionality of marriages between European men and Amerindian women.
