[cat] Els gliomes són el tipus més comú de tumor cerebral primari, i es caracteritzen per ser
altament invasius. Es classifiquen segons el grau de malignitat, sent del grau IV els
més malignes. Aquests tumors d'alt grau són els glioblastomes, un tipus de tumor molt
heterogeni que resisteix a tots els tractaments actuals, en part a causa de l'elevada
invasivitat. Aquest fet els converteix en un focus d'estudi important. L'adquisició de la
capacitat invasiva s'ha relacionat amb l'aparició de diverses mutacions genòmiques a
les cèl·lules dels glioblastomes que desregulen importants vies de senyalització
implicades en el control de la proliferació, creixement, migració i apoptosi.
Aproximadament un 80% dels glioblastomes contenen una senyalització aberrant dels
receptors tirosina quinasa (RTKs), ja sigui per amplificació o per mutacions en els gens
dels receptors. El present estudi es centra en analitzar els efectes de la senyalització
dels RTKs sobre la motilitat cel·lular i l'organització del citoesquelet d'actina en
cèl·lules primàries derivades de gliomes malignes. Per a observar realment dits
efectes s'han realitzat Wound Healing Assays per quantificar la motilitat cel·lular i
tècniques de microscòpia confocal per observar les diferències en el citoesquelet entre
cèl·lules primàries de glioma control i tractades amb inhibidors de RTK. Els resultats
obtinguts demostren que el tractament amb inhibidors de RTK provoca una reducció
de la motilitat cel·lular i un increment del nombre d'adhesions focals i fibres d'estrès
en el citoesquelet de les cèl·lules primàries, suggerint que els RTK són interessants
dianes terapèutiques per al tractament dels gliomes malignes. |

[eng] Gliomas are the most common type of primary brain tumor, and are characterized by
being highly invasive. They are classified according to the degree of malignancy, being
the grade IV the most malignant ones. These high-grade tumors are glioblastomas, a
type of heterogeneous tumor that resists all current treatments, partly because of the
high invasiveness. This makes them an important focus of study. The acquisition of
invasive capacity has been linked to the emergence of several genomic mutations in
the cells of glioblastomas that deregulated signaling pathways involved in the control
of proliferation, growth, migration and apoptosis. Approximately 80% of glioblastomas
contain an aberrant signaling of tyrosine kinase receptors (RTKs), either by
amplification or mutations in the genes of the receptors. This study focuses on
analyzing the effects of RTKs signaling on cell motility and organization of the actin
cytoskeleton in cells derived from primary malignant gliomas. To really see these
effects Wound Healing Assays were performed to quantify cell motility and confocal
microscopy techniques to observe the differences between the cytoskeleton of primary
glioma cells control and treated with RTK inhibitors. The results demonstrate that
treatment with RTK inhibitors causes a reduction in cell motility and an increase in the
number of focal adhesions and stress fibers in the cytoskeleton of primary cells,
suggesting that RTK are interesting therapeutic targets for the treatment of malignant
gliomas. |
