[cat] Les sirtuïnes són proteïnes desacetilases dependents de NAD+ que intervenen
regulant diversos processos cel·lulars com ara la inflamació, la senescència, la diferenciació i
el metabolisme cel·lular, a més del control de la funció mitocondrial i la resposta antioxidant.
En aquest estudi ens centram en la Sirtuïna 1 (SIRT1) i Sirtuïna 3 (SIRT3) que divergeixen de
localització subcel·lular i funcions principals. SIRT1 en concret, es troba en el nucli i en el
citoplasma. A més, té com a principals funcions la inhibició de les vies
apoptòtiques, angiogèniques i proliferatives, així com la reparació del DNA, entre altres. En
canvi, la SIRT3 es troba en gran part a la mitocòndria, on du a terme les seves funcions
regulant el metabolisme energètic, la supervivència cel·lular, el balanç de l'ATP, la maquinària
antioxidant, control de l'apoptosi i altres mecanismes que es donen en la mitocòndria.
Aquestes sirtuïnes estan involucrades en el desenvolupament del càncer de còlon, ja que
poden actuar com a protumorals o antitumorals presentant així un paper dual.
L'expressió d'aquestes proteïnes ha estat detectada per Western Blot, amb 23 mostres
provinents de tumors d'estadis I-IV de càncer colorectal. S'ha vist que hi ha una tendència a
la disminució en l'expressió d'ambdues sirtuïnes a mesura que progressa el tumor. Per tant,
els resultats suggereixen que les sirtuïnes a estadis més avançats del càncer actuen com a
supressors tumorals, de manera que la cèl·lula maligne disminueix la seva expressió per evitar
la seva acció antitumoral.
[eng] Sirtuins are NAD + dependent deacetylase proteins that regulates various cellular processes
such as inflammation, senescence, differentiation and cell metabolism, as well as control of
the mitochondrial function and the antioxidant response. In this study we focus on Sirtuin 1
(SIRT1) and Sirtuin 3 (SIRT3) that diverge from subcellular location and main functions. SIRT1
is found in the nucleus and the cytoplasm and has as main functions the inhibition of the
apoptotic, angiogenic and proliferative pathways, as well as the repair of DNA, among others.
In contrast, the majority of SIRT3 is found in the mitochondria, where regulates the energy
metabolism, cell survival, balance of ATP, antioxidant machinery, apoptosis pathways and
other mechanisms that occur in mitochondria.
These sirtuins are involved in the development of colon cancer, since they can act as
protumorals or antitumorals, presenting a dual role.
The expression of these proteins has been detected by Western Blot, in 23 samples from I-IV
stage tumors of colorectal cancer. The results shows that there is a tendency to decrease the
expression of both sirtuins as the tumor progresses. Therefore, the results suggest that the
sirtuins at more advanced stages of cancer act as tumor suppressors, so that the malignant
cell decreases its expression to prevent their antitumor action.
