[cat] L’osteoartritis és una malaltia reumatològica causada per una progressiva
degeneració del cartílag i l’òs subcondral que conformen les articulacions.
L’elevada prevalença a la població que sofreix osteoartritis, la converteix en un
focus d’investigació per a la recerca de noves teràpies que no sols intentin pal·liar
els símptomes si no que aconsegueixin una regeneració del teixit. Es postulen
teràpies basades en l’ús de cèl·lules mare mesenquimals, aquest treball es
centra específicament en les que provenen de cordó umbilical (hUC-MSC) per la
seva capacitat de regeneració tissular i de diferenciació en condròcits. Però l’ús
de teràpies cel·lulars presenta uns certs inconvenients com la possible activitat
tumorigènica i l’activació de respostes immunitàries causants de rebutjos. Per
aquests motius sorgeix una nova línia d’investigació entorn a les vesícules
extracel·lular (VEs). Les VEs són producte de les hUC-MSC i tenen la funció de
ser intermediaris i missatgers cel·lulars, amb l’avantatge de que al ser derivats
cel·lulars no presenten els inconvenients de les teràpies convencionals
esmentades. En aquest treball es persegueix avaluar l’ús de les VEs aïllades de
hUC-MSC com a possible tractament regeneratiu sobre un model in vitro
d’osteoartritis a partir de cèl·lules d’una línia condrogènica ATDC-5. Els
paràmetres avaluats tals com proves de citotoxicitat, d’activitat metabòlica, de
quantificació de col·lagen total i d’expressió gènica ens donen indicis de la
viabilitat del tractament i d’un cert grau de protecció i regeneració tissular. |

[eng] Osteoarthritis is a rheumatological disease caused by a progressive
degeneration of the cartilage and the subchondral bone that form the joints. The
high prevalence of population suffering from osteoarthritis makes it a research
focus in order to find new therapies that not only try to relieve symptoms but also
to achieve tissue regeneration. Therapies based on the use of mesenchymal
stem cells are postulated and this project focuses specifically on those that come
from umbilical cord (hUC-MSC) due to their capacity for tissue regeneration and
differentiation to chondrocytes. But the use of cellular therapies has certain
disadvantages such as possible tumorigenic activity and the activation of immune
responses that cause rejections. For these reasons a new line of research arises
around the extracellular vesicles (VEs). The VEs are product of the hUC-MSC
and have the function of being intermediaries and cellular messengers, with the
advantage that cellular derivatives do not present the drawbacks of the
conventional therapies as mentioned above. In this project we want to evaluate
the use of isolated VEs from hUC-MSC as a possible regenerative treatment on
an in vitro model of osteoarthritis using the ATDC-5 chondrogenic cell line. The
parameters evaluated such as cytotoxicity tests, metabolic activity, total collagen
quantification and gene expression point out the viability of the treatment giving
certain degree of protection and tissue regeneration. |
