[eng] Alzheimer's disease (AD) is a neurodegenerative disorder that represents the leading cause of dementia worldwide.
Prevalence of AD is increasing year after year, to the point that it is clearly becoming an epidemic. Despite the abundant
lines of research open to understand and fight the disease, to date, there is no treatment, and the last approved drugs for
this pathology date from the 90's. That is why there is a pressing need to delevop new compounds to treat the disease
effectively. LP226 is an hydroxylated derivative of docosahexaenoic acid (DHA) wich has been shown to be effective in
many models to combat AD. On the one hand, it has been shown that treatment with LP226 recovers normal cognitive
function in transgenic mouse and Drosophila melanongaster models. On the other hand, the study of the
neuropathological characteristics of the disease has shown that LP226 prevents the accumulation of Amyloid-β (Aβ) and
phosphorylation of Tau protein in animals, while preventing cell death against other toxic stimuli typical of this
neuropathy, as oligomers of Aβ and excito-toxicity mediated by N-methyl-D-aspartate (NMDA). In the present study,
LP226 is used as lead compound from wich a series of derivatives (Lp226-II, Lp226-III, Lp226-IV, Lp226-V, Lp226-VI)
were synthesized in order to achieve better stability, toxiciy and effectiveness profiles than mother molecule ones. From the
new molecules synthesized, chemical stability will be analyzed as well as toxicity and pharmacological efficacy against
neurotoxic stimuli in neuron hipocampal primary culture. In conclusión, the results of this work could lead to the acquisition
of new compounds with less toxicity and improved stability and effectiveness for the treatment of neuropathies such as AD.
[spa] La enfermedad de Alzheimer (EA) es un desorden neurodegenerativo que representa la primera causa de demencia en el
mundo. La prevalencia de EA va en aumento año tras año por lo cual es considerada como epidemia. Pese a las numerosas
líneas de investigación abiertas para comprender y combatir la enfermedad, a día de hoy no existe ningún tratamiento eficaz
y los últimos fármacos aprobados para esta patología datan de la década de los 90. Es por eso, que se hace patente, la
necesidad de desarrollar nuevos compuestos que sean efectivos para tratarla. LP226 es un derivado hidroxilado del ácido
docosahexaenoico (DHA) que ha mostrado efectividad en numerosos modelos para combatir la EA. Por un lado, se ha
demostrado que el tratamiento con LP226 recupera la función cognitiva normal en modelos transgénicos de ratones y
Drosophila melanongaster. Por otro lado, el estudio de las características neuropatológicas de la enfermedad ha mostrado
que LP226 previene la acumulación de β-amiloide (Aβ) y la fosforilación de la proteína tau en animales, mientras que
previene la muerte celular frente diferentes estímulos tóxicos propios de esta neuropatía, como oligómeros de Aβ y excitotoxicidad mediada por N-metil-D-aspartato (NMDA). En el presente estudio, LP226 se emplea como cabeza de serie en la
síntesis de nuevos derivados (Lp226-II, Lp226-III, Lp226-IV, Lp226-V, Lp226-VI) con la finalidad de conseguir perfiles de
estabilidad, toxicidad y de efectividad mejores a los de la molécula madre. A partir de las nuevas moléculas sintetizadas, se
analizará por un lado la estabilidad química y, por otro lado, la toxicidad y la eficacia farmacológica frente a estímulos
neurotóxicos propios de la EA en cultivos neuronales. En conclusión, los resultados de este trabajo podrían derivar en la
obtención de nuevos compuestos con mejor estabilidad, y mejores perfiles toxicológico y terapéutico, destinados al
tratamiento de neuropatías como la EA.
