[spa] El principal mecanismo de resistencia a antibióticos β-lactámicos en bacterias Gramnegativas es la producción de β-lactamasas, AmpC en el caso de Pseudomonas
aeruginosa. Se están buscando nuevas estrategias para combatir este mecanismo de
resistencia mediante el desarrollo de inhibidores de β-lactamasas que administradas
en combinación con β-lactámicos son capaces de restablecer su actividad frente a
aislados productores de β-lactamasas. Este estudio tiene el objetivo de averiguar la
posible existencia de otros mecanismos de resistencia a los β-lactámicos
independientes del mecanismo clásico, para predecir la emergencia de esos
mecanismos cuando se traten infecciones de P. aeruginosa con combinación de βlactámicos/inhibidores de β-lactamasas. Se utilizó la cepa de referencia PAO1 y sus
mutantes knock-out, PAO∆C (mutante deficiente en el gen ampC) y PAO∆G (mutante
deficiente en el gen ampG, que codifica una permeasa necasaria para activar la
expresión del gen ampC). Estas cepas fueron incubadas en medios de Mueller-Hinton
con concentraciones crecientes del antibiótico ceftazidima (β-lactámico) durante 7 días
para seleccionar mutantes al antibiótico por exposición prolongada al mismo. Se
estudió la resistencia a 13 agentes antispeudomónicos para determinar la posibilidad
de aparecer resistencia cruzadas. Se secuenció el genoma de estos mutantes y se
comparó con el genoma de la cepa salvaje con el fin de averiguar en qué genes se
habían producido las mutaciones y el papel que desempeñaban esos genes en la
resistencia al antibiótico. |

[eng] The main mechanism of resistance to β-lactam antibiotics in Gram-negative bacteria is
the production of β-lactamases, AmpC in the case of Pseudomonas aeruginosa. New
strategies are being searched to combat this mechanism of resistance through the
development of β-lactamase inhibitors, which administered in combination with βlactams, can restore their activity against β-lactamase-producers isolates. The purpose
of this study is to determine the possible existence of other β-lactam resistance
mechanisms independent of the classical mechanism, to predict the emergence of
these mechanisms when treating P. aeruginosa infections with a combination of βlactams/β- lactamasas inhibitors. We used the reference strain PAO1 and its knockout
mutants, PAOΔC (mutant deficient in the ampC gene) and PAOΔG (mutant deficient in
the ampG gene, which encodes a permease to activate the expression of the ampC
gene). These strains were incubated in Mueller-Hinton broth with increasing
concentrations of the antibiotic ceftazidime (β-lactam) for 7 days to select mutants to
the antibiotic for prolonged exposure to it. Resistance to 13 antipseudomonal agents
was studied to determine the possibility of cross-resistance. The genome of these
mutants was sequenced and compared with the genome of the wild strain to find out in
which genes the mutations had been produced and the role played by these genes in
the resistance to the antibiotic. |
