[eng] Xanthohumol (XN), isoxanthohumol (IXN) and 8-prenylnaringenin (8-PN) are hop-derived
prenylflavonoids and have been reported to exhibit anticancer properties in several types of cancer.
They present a great interest against colon cancer due to high exposure of these compounds in this
tissue. Metastatic SW-620 cell line was treated with doses ranging from 0.001 to 10 µM of XN, IXN or
8-PN in order to assess their effects on cell viability and mitochondrial function determined by Hoechst
33342, Amplex Red, MitoTracker® Green and Western blot. At low concentrations, these compounds
had no effect on assays carried out. At high concentration, XN and IXN led to a decrease in cell viability,
while the 8-PN had no effects. In addition, at 10 μM XN, it gave rise to an increase in ROS production
accompanied by a decrease in OXPHOS complexes and sirtuin expression levels. These results suggest
that XN or IXN in low dose and 8-PN do not alter the viability or mitochondrial function, whereas a high
dose of XN induce mitochondrial dysfunction in SW620 colon cancer lines. |
