[spa] La enfermedad periodontal se caracteriza por la infección e inflamación de las encías
que puede conducir a su destrucción, y en caso graves, a la degradación del hueso
alveolar y a la perdida de dientes. El objetivo primordial del tratamiento periodontal es
la regeneración de los tejidos blandos y duros del periodonto y la eliminación de la
infección. En este estudio, se realizó un cultivo celular 2D de fibroblastos gingivales
humanos, para posteriormente generar una herida sobre ellos e inducir una inflamación
mediante un tratamiento con lipopolisacárido (LPS), con el fin de simular las condiciones
que se dan en la periodontitis. Posteriormente, se seleccionaron y se aplicaron
diferentes geles orales comerciales con factores de regeneración y/o factores
antibacterianos; y se analizó la citotoxicidad que éstos presentaban, así como su efecto
regenerativo y cicatricial. Además, se evaluó la expresión de diferentes genes
relacionados con la proliferación y diferenciación de fibroblastos y el mantenimiento de
la matriz extracelular.
Solamente los geles que no presentaban factores antimicrobianos o contenían
concentraciones muy bajas, además de contener componentes regenerativos
producidos por las propias células, demostraron ser biocompatibles, mientras que los
otros geles provocaron la muerte de los fibroblastos, debido al fuerte efecto citotóxico
que presentaron. De entre todos los geles testados, Emdogain es el que mostró mejor
cierre de la herida y mayores niveles de genes relacionados con la producción y
organización de la matriz extracelular. |

[eng] Periodontal disease is characterized by the infection and inflammation of the gingiva
that may lead to its destruction, and in severe cases, to the degradation of the alveolar
bone and tooth loss. The primary objective of the periodontal treatment is the
regeneration of the soft and hard tissues of the periodontium, and removal of infection.
In this study, we performed a 2D cell culture of human gingival fibroblasts, to
subsequently generate a wound on them and induce inflammation by
lipopolysaccharide (LPS) treatment, in order to simulate periodontitis conditions.
Subsequently, we selected and applied different commercial oral gels with regeneration
factors and/or antibacterial factors; and we analyzed the cytotoxicity they presented, as well as their regenerative and cicatricial effect. In addition, we evaluated the expression
of different genes related to the proliferation and differentiation of fibroblasts and
maintenance of the extracellular matrix.
Only the gels that did not present antimicrobial factors or contained very low
concentrations, in addition to containing regenerative components produced by the
cells themselves, proved to biocompatible, while the other gels caused the death of the
fibroblasts, due to the strong cytotoxic effect that they presented. Of all the gels tested,
Emdogain showed the best wound closure and highest levels of genes related to the
production and organization of the extracellular matrix. |
