[cat] L'Alzheimer és una malaltia neurodegenerativa que es caracteritza per la formació de plaques senils i agregats neurofibril·lars amb la conseqüent aparició de deterior cognitiu progressiu, demència, pèrdua progressiva de la memòria i independència reduïda. Hi ha nombrosos factors que poden incrementar el risc de patir Alzheimer. Concretament, en l'actualitat s'estan estudiant els efectes a nivell neurològic de les exposicions prenatals, postnatals, i a l’edat adulta a disruptors endocrins, substàncies exògenes que causen alteracions en la funció endocrina generant efectes adversos en la salut d’un individu i/o a la seva descendència. Un dels disruptors més estudiants és el Bisfenol A (BPA), un compost químic present en plàstics que poden ser emprats en l’àmbit alimentari. L’objectiu d’aquest treball és, a partir d’una recerca bibliogràfica definida, dilucidar quins són els mecanismes pels quals el disruptor pot provocar alteracions neurològiques.
El desenvolupament del sistema nerviós central (SNC) comprèn el procés de neurulació i neurogènesi (tant a una edat primerenca com a l’adulta), i aquests processos es troben regulats per estrògens de forma que són susceptibles de ser alterats per BPA. Concretament en aquest treball es descriu l’efecte del BPA sobre una via de senyalització implicada a diferents nivells en el desenvolupament del SNC i que es veu regulada pels estrògens. Es tracta de la via de la proteïna quinasa activada per mitògens (Mitogen-Activated Protein Kinase, MAPK o MAPK/ERK).
Una exposició prenatal-postnatal a BPA pot tenir conseqüències a nivell de formació del SNC i neurogènesi mitjançant l’alteració de la via de la MAPK. Per altra banda, en quant a exposicions a l’edat adulta, la disrupció de la via pot predisposar a l’individu a presentar una patogènia molt similar a la de L’Alzheimer. Cal mencionar que els efectes d’aquestes exposicions a BPA seran diferents en funció del sexe a causa del dimorfisme sexual al cervell; i també en funció de l’edat.
[eng] Alzheimer's disease is a neurodegenerative disease characterized by the formation of senile and neurofibrillary plaques with the consequent onset of progressive cognitive impairment, dementia, progressive loss of memory and reduced independence. There are numerous factors that may increase the risk of this disease. Specifically, the neurological effects on an individual or his offspring caused by a prenatal, postnatal or adult exposure to endocrine disruptors, exogenous substances that cause alterations in the endocrine function, are being studied. One of the most studied disruptor is Bisphenol A (BPA), a chemical compound present in plastics that can be used in the food field. The objective of this work is, based on a defined bibliographic research, to determine the mechanisms by which the disruptor can cause neurological disorders.
The central nervous system (CNS) development includes neurulation and neurogenesis (at an early age and adulthood), and these are regulated by estrogens, so they are susceptible to being altered by BPA. Specifically, in this work, the BPA effect on a signaling pathway involved at different levels in the development of the CNS and which is regulated by estrogens, is described. This is the mitogen-activated protein kinase (Mitogen-Activated Protein Kinase, MAPK or MAPK / ERK) pathway.
A prenatal-postnatal exposure to BPA may have consequences for the CNS formation and neurogenesis by altering MAPK's pathway. On the other hand, an adulthood exposure, by the same way, can predispose to present a similar Alzheimer’s disease pathogenesis. It should be mentioned that the BPA exposure effects will be different depending on sex because of the sexual dimorphism in the brain; and on the age.
