[spa] Algunas patologías crónicas con gran prevalencia entre la población como la obesidad comienzan a ser
desarrolladas desde etapas muy tempranas en el desarrollo humano. Se ha visto que alteraciones en el
estado nutricional de la madre favorecen el riesgo de aparición de dichas patologías en lo que se conoce
como “programación metabólica temprana”. Los miARNs han surgido como reguladores esenciales en
dicho proceso. Estas moléculas se ven alteradas en la leche materna en un estado obesogénico per se o
inducido por una dieta hipercalórica.
El objetivo del presente trabajo de fin de grado es analizar los efectos de un miARN específico alterado en
la leche materna por un ambiente obesogénico, el miR-200a, sobre la expresión de algunos genes
relevantes de la vía insulínica y el metabolismo lipídico en un cultivo celular de hepatocitos (HepG2).
Para ello, en primer lugar, se pretende validar la técnica de transfección in vitro mediante controles
positivos del mimético miR-1 y el inhibidor del miARN let-7c endógeno. Una vez verificado, se analiza la
expresión de los genes INSR, IRS1 y FASN tras la transfección in vitro con un mimético e inhibidor
específico del miR-200a. Además, se pretende corroborar si el IRS1, diana predictiva del miR-200a según
Target Scan es una diana directa o no.
Los resultados muestran un claro efecto del mimético del miR-200a sobre el receptor de insulina (INSR),
incrementando sus niveles. En cambio, los resultados obtenidos para la expresión del IRS1 y FASN no son
concluyentes, aunque se muestra una tendencia al incremento de la expresión de FASN, que podría ser
efecto directo de la presencia del mimético del miR-200a o indirecto por el incremento de INSR. Por
último, IRS1 no se ha podido validar como diana del miR-200a. |

[eng] Some chronic pathologies with high prevalence among the population such as obesity begin to be developed
from very early stages in human development. It has been seen that alterations in the nutritional state of the
mother favor the risk of appearance of such pathologies in what is known as “early metabolic programming”.
miRNAs have emerged as essential regulators in this process. These molecules are altered in breast milk in an
obesogenic state per se or induced by a hypercaloric diet.
The aim of this end-of-grade work is to analyze the effects of a specific miRNA altered in breast milk by an
obesogenic environment, miR-200a, on the expression of some relevant genes of the insulinic pathway and
lipid metabolism in a cell-hepatocytic culture (HepG2).
In order to do this, the first step is to validate the in vitro transfection technique by means of positive controls
of the miR-1 mimetic and the endogenous miRNA let-7c inhibitor. Once verified, the expression of the INSR,
IRS1 and FASN genes after in vitro transfection is analyzed with a miR-200a specific mimetic and inhibitor.
In addition, it is intended to corroborate whether the IRS1, predictive target of miR-200a according to Target
Scan is a direct target or not.
The results show a clear effect of the miR-200a mimic on the insulin receptor (INSR), increasing its levels. On
the other hand, the results obtained for the expression of IRS1 and FASN are not conclusive, although there is
a tendency to increase the expression of FASN, which could be a direct effect of miR-200a mimic or mediated
by the increase of INSR. Finally, IRS1 could not be validated as a target of miR-200a. |
