[spa] Yersinia enterocolitica modula la expresión de sus factores de virulencia para adaptarse
al huésped. En este trabajo hemos estudiamos la regulación de las 3 aciltransferasas
msbB, htrB y lpxP responsables de la acilación del lípido A por la temperatura y su
función en la patogénesis de Y. enterocolitica serotipo O:8. Los resultados mostraron
que Y. enterocolitica cambia la acilación del lípido A según la temperatura. Cuando la
bacteria crece a 21ºC expresa un lípido A principalmente hexaacilado, formado por un
lipido IVA modificado con los grupos C12 and C16:1. Estos grupos son añadidos por las
aciltransferasas MsbB y LpxP, respectivamente. A 37ºC un lípido A triacilado
modificado con un grupo C14 es la forma principal. Este grupo es añadido por la
aciltransferasa HtrB. Estudios transcripcionales mostraron que la expresión de las 3
aciltransferasas está regulada por la temperatura: msbB y lpxP se expresan a 21ºC
mientras que htrB se expresa 37º. Por otro lado, se mostró que el grado de acilación del
lípido A afecta a la expresión de inv, yplA, flhDC y el antígeno O, así como a la
resistencia de la bacteria frente a los péptidos antimicrobianos. Para terminar, los
experimentos realizados con ratones indicaron que HtrB es esencial para la virulencia de Y.
enterocolitica serotipo O:8. |

[eng] Yersinia enterocolitica modulate the expression of their virulence factors to adapt to the
host environment. In this work we have studied the regulation by temperature of the 3
late acyltransferases msbB, htrB and lpxP responsible for lipid A acylation and their role
in Y. enterocolitica serotype O:8 pathogenesis. The results showed that Y. enterocolitica
change the acylation of lipid A in response to temperature. The lipid A expressed by
bacteria grown at 21ºC was predominantly hexaacylated, consisting of a lipid IVA
modified with C12 and C16:1 acylgroups, which are added by msbB and lpxP
acyltransferases, respectively. At 37ºC the predominant form was a triacid lipid A
modified with C14, a acylgroup added by htrB acyltransferase. Transcriptional analysis
revealed that the expression of the acyltransferases msbB, htrB and lpxP was temperatureregulated: msbB and lpxP genes were expressed at 21ºC whereas htrB gene was expressed
at 37º. On the other hand, we demonstrated that the status of lipid A acylation affects the
expression of other Y. enterocolitica virulence factors: inv, yplA, flhDC and O-antigen.
Besides, this status affects the resistance of Yersinia to antimicrobial peptides. To finish,
animal experiment showed that HtrB acyltransferase is essential for full virulence of Y.
enterocolitica serotype O:8. |
