[cat] Les plaquetes juguen un paper fonamental en la cicatrització de ferides. Durant una sèrie d’anys, es tenia per establert que els efectes de les plaquetes eren mediats principalment per factors de creixement que alliberaven al medi extracel·lular, d’aquí que el lisat de plaquetes (LP) s’hagi consolidat com a teràpia per al tractament d’úlceres epitelials, un tipus de ferida crònica. No obstant això, nombroses evidències en la actualitat apunten a que els principals intermediaris de les funcions de les plaquetes serien les vesícules extracel·lulars (VEs) que en deriven d’elles (P-VEs). Per tant, en el present treball ha estat d’interès investigar l’ús potencial de les P-VEs com a tractament per al tancament de ferides. En el present treball s’han aïllat P-VEs per cromatografia d’exclusió de mida (SEC), i s’han caracteritzat mitjançant anàlisi de rastreig de nanopartícules (NTA), microscopia electrònica de transmissió (TEM) i Western Blot. Les P-VEs aïllades es varen utilitzar per al tractament de ferides induïdes en cultius cel·lulars 2D de fibroblasts gingivals humans immortalitzats, i es varen comparar amb un tractament basat en LP i amb un grup control sense tractament. Els canvis en el tancament de la ferida es varen avaluar a les 3 i 24 h d’incubació amb els respectius tractaments, demostrant diferències significatives amb el tractament de P-VEs després de 24 h respecte al grup control. A més a més, vàrem demostrar, per una banda, com aquest tractament no influïa sobre la viabilitat de les cèl·lules i, per altra banda, com l’efecte citotòxic resultant podia ser menyspreable. Així doncs, convindria seguir investigant en un futur el potencial terapèutic de les P-VEs com a tractament d’úlceres epitelials, per la qual cosa seria necessari emprar models més representatius del teixit humà, com ara els cultius cel·lulars en tres dimensions (3D).
[eng] Platelets play a fundamental role in wound healing. For years, it has been considered that the effects of platelets were mainly mediated by the growth factors that were released into the extracellular environment, hence, the platelet lysate (PL) has been used as a therapy for the treatment of epithelial ulcers, a type of chronic wound. However, present evidences suggest that the main intermediaries of platelet functions would be the extracellular vesicles (VEs) that derive from them (P-VEs). Therefore, in the present work it has been of interest to investigate the potential use of P-VEs as a treatment for wound closure. In the present work, P-VEs have been isolated by size exclusion chromatography (SEC), and characterized by nanoparticle tracking analysis (NTA), transmission electron microscopy (TEM) and Western Blot. The isolated P-VEs were used to treat induced wounds in 2D cell cultures of immortalized human gingival fibroblasts, compared to a treatment based on LP and to an untreated control group. The changes in wound closure were evaluated after 3 and 24 hours of treatment, showing significant differences on wound closure after 24 h the P-VEs treatment compared to the control group. Furthermore, we demonstrated, on the one hand, how this treatment did not influence the viability of the cells and, on the other hand, how the resulting cytotoxic effect could be negligible. Therefore, it would be advisable to continue investigating in the future the therapeutic potential of P-VEs as a treatment for epithelial ulcers, for which it would be necessary to use more representative models of human tissue, such as three-dimensional (3D) cell cultures.
