[cat] L’obesitat és una malaltia crònica, d’origen multifactorial i d’elevada prevalença. Es caracteritza per un increment patològic del greix corporal. Estudis recents han posat de manifest que l’obesitat juntament amb l’estrès afecten a la regulació de l’eix hipotàlem-hipòfisi-adrenal, provocant una hipersecreció dels glucocorticoides. El cortisol, glucocorticoide amb funció catabòlica, està present a la llet materna i per tant, pot modular el metabolisme infantil. A més, s’ha observat l’existència d’una interconnexió entre els elevats nivells de cortisol i el síndrome metabòlic. Per aquest motiu, l’objectiu principal del present treball és estudiar si la ingesta d’una dieta obesògena associada a l’obesitat materna en rates pot afectar els nivells de cortisol de la llet materna. L’estudi experimental s’ha dut a terme amb rates Wistar femelles de dos grups experimentals, les rates del grup control i les rates del grup cafeteria, aquestes darreres han estat alimentades amb una dieta rica amb greixos i sucres, des del primer dia de lactància. Les mostres s’han obtingut a dia 5, 10 i 15 de lactància. Després d’analitzar els resultats, s’ha observat que les rates del grup cafeteria tenen un pes corporal i un percentatge de massa magra inferior a les rates del grup control. En canvi, el percentatge de massa grassa i els nivells de glucosa circulant són significativament superiors a les rates del grup cafeteria, en comparació amb el grup control. Respecte als nivells de cortisol a la llet, en primer lloc, es va fer una recerca dels possibles assajos comercials disponibles per quantificar l’esteroide. D’aquests s’ha seleccionat un kit ELISA de tipus competitiu. Els resultats obtinguts indiquen que els nivells de cortisol a llet materna són significativament inferiors a les rates del grup cafeteria respecte a les del grup control. Per tant, el consum d’una dieta obesògena té efectes rellevants sobre el metabolisme i la regulació hormonal en rates.
[eng] Obesity is a chronic disease of multifactorial origin and high prevalence. It is characterized by a pathological increase in body fat. Recent studies have shown that obesity along with stress affect the regulation of the hypothalamic-pituitary-adrenal axis, causing hypersecretion of glucocorticoids. Cortisol, a glucocorticoid with catabolic function, is present in breast milk and therefore can modulate infant metabolism. In addition, the existence of an interconnection between high cortisol levels and metabolic syndrome has been shown. For this reason, the main objective of the present study is to study whether the intake of an obese diet associated with maternal obesity in rats can affect the cortisol levels of breast milk. The experimental study was carried out with female Wistar rats from two experimental groups, the rats in the control group and the rats in the cafeteria group, the latter being fed a diet rich in fats and sugars, from day one of breast-feeding. Samples were obtained on days 5, 10, and 15 of breast-feeding. After analysis of the results, it has been observed that the cafeteria rats have a body weight and a percentage of lean mass below the rats in the control group. In contrast, the percentage of fat mass and circulating glucose levels are significantly higher in cafeteria rats than in the control group. Regarding to cortisol levels in breast milk, first, an investigation was carried out into the possible commercial trials available to quantify the steroid. A competitive ELISA kit has been selected from these. Results indicate that levels of cortisol in breast milk are significantly lower in the cafeteria group rats than in the control group rats. In conclusion, the intake of an obesogenic diet has relevant effects on metabolism and hormonal regulation of rats.
