[spa] El cáncer de colon representa una de las principales causas de morbilidad y mortalidad a nivel mundial, con una elevada mortalidad atribuible principalmente a la metástasis. El proceso metastásico implica varias etapas donde las células cancerosas modulan su metabolismo dependiendo de los requerimientos de cada estadio. En los últimos años, se ha incluido como característica distintiva del cáncer, los cambios en el metabolismo energético de la célula cancerosa, donde la disfunción mitocondrial puede provocar estrés oxidativo, el cual se ha asociado a la oncogénesis y la progresión metastásica.
Esta tesis ha tenido como objetivo general la identificación de posibles biomarcadores de pronóstico, relacionados con el metabolismo energético y el estrés oxidativo en el tejido tumoral y en el tejido adyacente no tumoral de pacientes en estadios avanzados III y IV de cáncer de colon. Para ello, se analizaron las adaptaciones relacionadas con la función mitocondrial y el estrés oxidativo en biopsias de pacientes en estadio pre-metastásico (III) y metastásico (IV) de cáncer de colon, comparando tejido tumoral y el tejido adyacente no tumoral del mismo paciente. Los resultados obtenidos muestran que el tejido tumoral pre-metastásico presenta un perfil metabólico oxidante similar al tejido adyacente no tumoral del mismo estadio, con tendencia a promover la función mitocondrial al alcanzar la metástasis. Además, la función mitocondrial presentó diferencias entre los estadios avanzados de cáncer de colon, siendo mayor en el estadio metastásico, aunque se observaron menores niveles proteicos de los complejos OXPHOS en el tejido tumoral del estadio IV. Por otra parte, los tejidos tumorales de ambos estadios presentaron unos niveles de estrés oxidativo muy similares. Sin embargo, en las muestras de tejido adyacente no tumoral de ambos estadios se encontraron las mayores diferencias relacionadas con el estrés oxidativo, mostrando el tejido adyacente no tumoral del estadio metastásico niveles proteicos y de actividades enzimáticas superiores de las proteínas antioxidantes analizadas, reflejando así la respuesta de este tejido frente a un elevado nivel de estrés oxidativo del tejido tumoral.
En conclusión, la presente tesis contribuye a profundizar en el conocimiento del metabolismo energético en estadios avanzados de cáncer de colon, permitiendo identificar, en el metabolismo oxidativo y la función mitocondrial del tejido tumoral, potenciales biomarcadores de pronóstico y evolución del cáncer de colon. Además, las características relacionadas con el estrés oxidativo del tejido adyacente no tumoral podrían también ser evaluadas como biomarcadores de pronóstico de utilidad clínica en pacientes de estadios avanzados de cáncer de colon.

En els últims anys, s'ha inclòs com a característica distintiva del càncer, els canvis en el metabolisme energètic de la cèl·lula cancerosa, on la disfunció mitocondrial pot provocar estrès oxidatiu, el qual s'ha associat a la oncogènesis i la progressió metastàtic.
Aquesta tesi ha tingut com a objectiu general la identificació de possibles biomarcadors pronòstic, relacionats amb el metabolisme energètic i l'estrès oxidatiu en el teixit tumoral i en el teixit adjacent no tumoral de pacients en estadis avançats III i IV de càncer de còlon. Per això, es varen analitzar les adaptacions relacionades amb la funció mitocondrial i l'estrès oxidatiu en biòpsies de pacients en estadi pre-metastàtic (III) i metastàtic (IV) de càncer de còlon, comparant teixit tumoral i teixit adjacent no tumoral del mateix pacient. Els resultats obtinguts mostren que el teixit tumoral pre-metastàtic presenta un perfil metabòlic oxidant similar al teixit adjacent no tumoral del mateix estadi, amb tendència a promoure la funció mitocondrial en aconseguir la metàstasi. A més, la funció mitocondrial va presentar diferències entre els estadis avançats de càncer de còlon, sent major en l’estadi metastàtic, encara que es varen observar menors nivells proteics dels complexos OXPHOS en el teixit tumoral de l'estadi IV. D'altra banda, els teixits tumorals de tots dos estadis varen presentar uns nivells d'estrès oxidatiu molt similars. No obstant això, en les mostres de teixit adjacent no tumoral de tots dos estadis es varem trobar les majors diferències relacionades amb l'estrès oxidatiu, mostrant el teixit adjacent no tumoral de l'estadi metastàtic nivells proteics i d'activitats enzimàtiques superiors de les proteïnes antioxidants analitzades, reflectint així la resposta d'aquest teixit enfront d'un elevat nivell d'estrès oxidatiu del teixit tumoral.
En conclusió, la present tesi contribueix a aprofundir en el coneixement del metabolisme energètic en estadis avançats de càncer de còlon, permetent identificar, en el metabolisme oxidatiu i la funció mitocondrial del teixit tumoral, potencials biomarcadors de pronòstic i evolució del càncer còlon. A més, les característiques relacionades amb l'estrès oxidatiu del teixit adjacent no tumoral podrien també ser avaluades com biomarcadors pronòstic d'utilitat clínica en pacients d'estadis avançats de càncer de còlon.

The metastatic process involves several stages where the cancer cells modulate their metabolism depending on the requirements of each stage. In recent years, changes in the cancer cell's energy metabolism have been included as a distinctive feature of cancer, where mitochondrial dysfunction can cause oxidative stress, which has been associated with tumorigenesis and metastatic progression.
The general objective of this thesis has been to identify possible prognostic biomarkers related to energy metabolism and oxidative stress in tumor tissue and in adjacent non-tumor tissue of patients in advanced stages III and IV of colon cancer. To this end, adaptations related to mitochondrial function and oxidative stress in biopsies from patients in pre-metastatic (III) and metastatic (IV) stages of colon cancer were analyzed, comparing tumor tissue and adjacent non-tumor tissue from the same patient. The results obtained show that pre-metastatic tumor tissue presents an oxidant metabolic profile similar to the adjacent non-tumor tissue of the same stage, with a tendency to promote mitochondrial function when metastasis is reached. In addition, mitochondrial function showed differences between advanced stages of colon cancer, being higher in the metastatic stage, although lower protein levels of OXPHOS complexes were observed in stage IV tumor tissue. On the other hand, the tumor tissues of both stages showed very similar levels of oxidative stress. However, the greatest differences related to oxidative stress were found between the non-tumor adjacent tissue samples of both stages, with the adjacent non-tumor tissue of the metastatic stage showing higher protein levels and enzyme activities of the antioxidant proteins analyzed, thus reflecting the response of this tissue to a high level of oxidative stress in the tumor tissue.
In conclusion, the present thesis contributes to deepen the knowledge of energy metabolism in advanced stages of colon cancer, allowing the identification, in the oxidative metabolism and mitochondrial function of the tumor tissue, potential biomarkers of prognosis and evolution of colon cancer. In addition, characteristics related to oxidative stress of adjacent non-tumor tissue could also be evaluated as prognostic biomarkers of clinical utility in patients with advanced stages of colon cancer.
