[spa] El glioblastoma multiforme (GMB) es un tipo de tumor del Sistema Nervioso Central que se origina por una proliferación en astrocitos (astrocitoma), considerándose es el tipo de astrocitoma más severo (grado 4). FoxM1 es un factor de transcripción que regula la expresión de genes relacionados con vías de señalización que conducen a un crecimiento celular (progresión del ciclo), división celular, proliferación, angiogénesis y supervivencia celular. Se ha comprobado que FoxM1 puede encontrarse sobreexpresado en muchos tipos de tumores, incluido GMB. Por otro lado, otro gen importante en la regulación del ciclo celular es FoxO3a. Este factor de transcripción regula procesos como proliferación, apoptosis, metabolismo y longevidad. La regulación de FoxO3a depende de Akt, proteína que fosforila a FoxO3a e inhibe su actividad transcripcional. En este estudio, se trabaja con Thiostrepton (agente tiazolado), el cual inhibe FoxM1, y con Trifluoperazina, fármaco que activa FoxO3a. El estudio se ha realizado en dos líneas celulares de GMB: U87 MG y LN229. Se han realizado ensayos de viabilidad en ambas líneas con los dos fármacos para obtener la IC50 de ambas líneas, se ha comprobado si Thiostrepton (TS) puede tener efecto sinérgico con la quimioterapia convencional de GBM (Temozolamida) mediante ensayos de viabilidad y, por último, se ha analizado la actividad caspasa de ambas líneas después del tratamiento con Thiostrepton, Temozolamida y la combinación de ambos.
[cat] Glioblastoma multiforme (GMB) is one the most severe cancers of the Central Nervous System and the most severe type of astrocytome (4th grade). FoxM1 is a transcription factor which regulates genes directly related with proliferation and cell survival. Overexpression of FoxM1 is a common feature of different types of cancer such as GMB. FoxO3a is another important gene for cell regulation. Apoptosis is one of the most important events regulated by FoxO3a. FoxO3a is downregulated by the PI3K pathway via Akt-dependent phosphorylation. In this work, we wanted to analyse the effect of two drugs, Thiostrepton which is a thiazole agent and FoxM1 inhibitor, and trifluoperazine which is a FoxO3a activator, in two GMB cell lines: U87 MG and LN229. Viability assays have been conducted in both cell lines with Thiostrepton and Trifluoperazine to obtain the IC50. Furthermore, we have analysed the synergistic effect of Thiostrepton together with the conventional chemotherapeutic agent temozolamide (TMZ) by viability essays. Finally, we have analyzed the caspase activity at different concentrations of Thiostrepton, Temozolamide and the combination.
